Large and complex proteins are usually synthesized in the rough endoplasmic reticulum (ER) where there are ribosomes attached to the membrane. The ribosomes translate the mRNA into the protein, which is then translocated across the ER membrane and into the ER lumen where it can be folded and modified before being transported to its final destination within or outside the cell.
