Topoisomerases are enzymes that help relieve tension in the double helix structure of DNA by cutting one or both strands of DNA, changing its topology (twisting), and then re-joining the strands. DNA gyrase is one of the most well-known topoisomerases and is responsible for introducing negative supercoils into the DNA during DNA replication in bacteria. DNA ligase seals gaps in the sugar-phosphate backbone of double-stranded DNA that are left during the normal course of DNA replication, repair, and recombination. DNA polymerase I and III are involved in DNA replication, but not in topoisomerization.
