Hydrophobic hormones, such as peptides and amino acids, bind externally to the plasma membrane receptors that activate protein kinases directly or that operate through second-messenger systems such as cAMP. Steroids, which are lipophilic, pass through a target cell's plasma membrane and bind to intracellular receptor proteins. The hormone-receptor complex then binds to the hormone response element of the target gene.
