The F plasmid is composed of circular double-stranded DNA. However, for transfer by conjugation, The F plasmid is nicked at the origin of transfer site. Then, the nicked strand is transferred to the recipient cell as a linear single-stranded molecule while the intact circular strand remains in the donor cell. Once in the recipient cell, the linear DNA is rejoined. A second strand of DNA is then synthesized in both cells.
