An amino acid will only be in its fully protonated form (NH3+ and COO-) at low pH values. As the pH increases, the amino acid will lose protons and eventually become a zwitterion with a net charge of zero at its isoelectric point (pI). At pH values above the pI, the amino acid will have a net negative charge due to the deprotonated carboxyl group, and at pH values below the pI, the amino acid will have a net positive charge due to the protonated amino group. Therefore, an amino acid will have a pH less than its isoelectric point.
