Fatty acyl-CoA cannot cross the inner mitochondrial membrane directly. It must first be converted to fatty acyl-carnitine by the enzyme carnitine palmitoyltransferase I (CPT I) and then transported across the inner mitochondrial membrane by the carnitine-acylcarnitine translocase. Once inside the mitochondria, the fatty acyl-carnitine is converted back to fatty acyl-CoA by the enzyme carnitine palmitoyltransferase II (CPT II) so that it can undergo beta-oxidation.
