High-resolution banding is achieved by staining the chromosomes before maximal condensation. This allows for better visualization of the distinct banding patterns on the chromosomes. Increasing the concentration of Giemsa or staining the chromosomes longer may lead to over-staining and loss of resolution. Staining at low temperatures may slow down the staining process but does not necessarily improve resolution.
