Progesterone receptors can indeed bind to DNA in the absence of progesterone, though their activity is significantly enhanced in the presence of the hormone. They can also be activated independently of progesterone through phosphorylation. Additionally, in the absence of progesterone, these receptors are associated with heat shock proteins in the cytoplasm, which keep them in an inactive state. Upon binding progesterone, they undergo a conformational change, dissociate from the heat shock proteins, and can then enter the nucleus to exert their effects, facilitated by a nuclear localization signal they possess.
