Nucleotide sequencing provides the most precise and detailed information about the RNA sequence, allowing for an exact comparison to determine if two RNAs are identical.  In order to accurately determine the identity of any given RNA it is necessary to determine the nucleotide sequences of the encoding gene or to generate cDNA clones of the RNAs and compare their nucleotide sequences.
