Competitive inhibitors bind to the active site of the enzyme, preventing the substrate from binding. To overcome this, increasing the concentration of the correct substrate in the reaction would increase the chances of the substrate binding to the enzyme rather than the inhibitor. Adding a noncompetitive inhibitor or an allosteric activator would not affect the binding of the competitive inhibitor, and adding a cofactor may not necessarily overcome the inhibition caused by the competitive inhibitor. Hence, increasing the concentration of the correct substrate is the best way to overcome the problem caused by a competitive inhibitor.
