A polar mutation is a mutation in one gene that decreases expression of a downstream gene in an operon. As a consequence, mutations in the last gene in an operon are not usually polar as there is no downstream gene for them to affect. Polar mutations often increase the noncoding distance between genes, which can adversely affect transcription or translation of the downstream genes. Genes affected by polar mutations are not usually rescued by supplying a copy on a plasmid.
