Gram-negative bacterial cells do not have a thick peptidoglycan layer with teichoic acid cross-linkages; therefore,when the decolorizing agent (acetone,alcohol,or a combination of both)is added the crystal violet-iodine complex is washed out.The bacteria will be colorless at this point and when counterstained with safranin will be red/pink in color.
